環境汚染物質の簡易・高感度計測を目指し、携帯型バイオセンサー装置を開発した。本装置は、使い捨て検出容器を装着後、外部パソコン制御にて全自動で簡単に測定を行うことができる。試料中の化学物質は、加えた蛍光性抗体と化学物質との抗原抗体反応速度を検出容器内で蛍光経時変化として計測することでモニタリングされる。本装置を用いてビタミンH、ポリ塩化ビフェニル、ダイオキシン、女性ホルモンなどの検出が可能であった。

概要 (英文)

Environmental contamination by artificial chemicals such as endocrine disruptors and dioxins has been focused in recent years. Such demands to sense these compounds existing at low concentration in environment created sensors using biological agents for their detection. However, any immunoassay instruments have not been available for environmental testing of the contaminated chemicals at present. Recently, we developed a high sensitive antibody based immunoassay. The purpose of this research is to develop an automated portable instrument capable of monitoring the chemicals with high sensitivity with our antibody based assay system. The portable instrument (25 cm Height×20 cm Wide × 20cm Depth) is one of the smallest among the commercially available equipments, which can automatically perform the assay under motion control by personal computer. The instrument can automatically mix the sample with the necessary reagents such as fluorescently labeled antibodies, control antigens and buffer. Then, the prepared mixture is flown thorough the fiber containing excess amount of the immobilized capturing reagents as a binding partner of the antibody in the disposable cell. The free antibody in the mixture was captured onto the fiber during the flow. The fluorescence from the captured antibody could be continuously measured through the surface lense on the cell. A time dependent changes of the fluorescence signal during the flow (slopes) were proportional to the concentrations of the free antibody in the sample mixtures. The free antibody concentrations must be related to the analyte concentrations. The slope is therefore the function of the analyte concentrations. In this regard, the instrument could monitor the analyte at near real time as a slope of the fluorescence signal. The performance of the instrument is tested with the biotin monitoring. A series of the samples containing biotin at the eight different concentrations was continuously flown in the instrument for 20 minutes. During the measurement, a time-dependent change of the signal was continuously recorded as a function of the biotin concentrations in the samples. It was possible to determine the biotin concentrations from 1ppb to 60ppb. This portable instrument serves not only as the convenient assay platform with high sensitivity but also as a semi-continuous monitoring method for the other target analytes. The dynamic range of the female formone (estradiol), dioxin(2,3,4,7,8-pentachloro dibenzofrane) and coplanar polychlorinated biphenyl(PCB#169) was 1ppt to 100ppt, 400ppt to 40ppb and 40ppt to 6ppb, respectively.
